He is best known for his work in the field of medical genetics, particularly in the area of metabolic diseases. Nyhan graduated from the University of Minnesota Medical School in 1951 and completed his residency in pediatrics at the University of Minnesota in 1954. He then went on to become a professor of pediatrics at the University of California, San Diego, where he served as the head of the Division of Medical Genetics from 1965 to 1991. Nyhan has made numerous contributions to the field of medical genetics, including the discovery of the genetic basis of several metabolic diseases, such as Lesch-Nyhan syndrome, and the development of a number of treatments for these diseases. He has also been involved in the development of genetic testing for a variety of conditions, including cystic fibrosis and Tay-Sachs disease. Nyhan has received numerous awards and honors for his work, including the National Medal of Science in 1991 and the Albert Lasker Award for Clinical Medical Research in 1992.
